研究概述
在《物種起源》中,達(dá)爾文提出"適者生存"的重要概念。"適者生存"的關(guān)鍵是成功應(yīng)對(duì)生存威脅。微觀尺度上的生存威脅主要是病原微生物。它們可以入侵人和動(dòng)物的消化道和呼吸道,引發(fā)一系列病理性的防御反應(yīng)(如惡心和嘔吐)。宏觀尺度上的生存威脅主要是天敵。在面臨天敵威脅時(shí),人和動(dòng)物表現(xiàn)出一系列生理性防御反應(yīng)(如戰(zhàn)斗和逃跑)。大腦如何感知這些不同尺度的生存威脅并啟動(dòng)相應(yīng)的防御反應(yīng),是神經(jīng)科學(xué)領(lǐng)域一個(gè)重要的科學(xué)問(wèn)題。我們實(shí)驗(yàn)室以小鼠為動(dòng)物模型,在微觀尺度上,研究了大腦啟動(dòng)惡心和嘔吐的神經(jīng)機(jī)制;在宏觀尺度上,研究了大腦啟動(dòng)戰(zhàn)斗和逃跑的神經(jīng)機(jī)制。
研究?jī)?nèi)容-1: 大腦感知病原入侵啟動(dòng)防御反應(yīng)的神經(jīng)機(jī)制
食物中毒引發(fā)的惡心和嘔吐是機(jī)體自我保護(hù)的防御反應(yīng),對(duì)生存具有重要的意義。惡心和嘔吐也是癌癥化療的主要副作用,具有重要的臨床意義。在過(guò)去三十年里,惡心和嘔吐的機(jī)制研究一直舉步維艱,因?yàn)轭I(lǐng)域內(nèi)缺乏合適的動(dòng)物模型和研究范式。
2022年11月1日,我們?cè)贑ell上發(fā)表了題為The gut-to-brain axis for toxin-induced defensive responses的研究論文。該研究建立了以小鼠為動(dòng)物模型研究惡心和嘔吐反應(yīng)的新范式,發(fā)現(xiàn)腸道報(bào)告病原入侵的“情報(bào)員”細(xì)胞,迷走神經(jīng)傳輸情報(bào)的“線人”細(xì)胞,以及腦內(nèi)負(fù)責(zé)啟動(dòng)惡心和嘔吐反應(yīng)的“開(kāi)關(guān)”細(xì)胞。在分子細(xì)胞層面上,揭示免疫反應(yīng)和5-羥色胺信號(hào)在引發(fā)惡心和嘔吐中的重要功能,證明化療藥物是通過(guò)“綁架”這些分子細(xì)胞機(jī)制引發(fā)惡心和嘔吐的副作用。研究還發(fā)現(xiàn),這些分子細(xì)胞和神經(jīng)環(huán)路機(jī)制也參與了化療藥物(doxorubicin)引發(fā)的“惡心-嘔吐”副作用。這一發(fā)現(xiàn)暗示,化療藥物引發(fā)的“惡心-嘔吐”副作用,可能是通過(guò)綁架進(jìn)化上保守的“食物中毒”機(jī)制得以實(shí)現(xiàn)。
同期的 Cell 配發(fā)了專家評(píng)論文章,對(duì)這一工作進(jìn)行了詳細(xì)介紹。研究成果還被Nature Reviews Neuroscience以Research Highlight的形式進(jìn)行了報(bào)道。紐約時(shí)報(bào)也對(duì)這一研究成果進(jìn)行了采訪和報(bào)道。
研究?jī)?nèi)容-2:大腦感知天敵威脅啟動(dòng)“戰(zhàn)斗-逃跑”反應(yīng)的神經(jīng)機(jī)制
人和動(dòng)物在面臨生存威脅(例如天敵)時(shí),可以表現(xiàn)出“戰(zhàn)斗-逃跑”行為反應(yīng),《水滸傳》中的武松打虎便是這一過(guò)程的生動(dòng)范例。在某些人群中,過(guò)度或反復(fù)的“戰(zhàn)斗-逃跑”行為反應(yīng)引發(fā)一類稱為創(chuàng)傷后壓力應(yīng)激障礙(PTSD)的精神疾病。要揭示一類疾病的發(fā)病機(jī)制,往往需要從源頭入手分析。解析引發(fā)“戰(zhàn)斗”和“逃跑”行為反應(yīng)的關(guān)鍵神經(jīng)環(huán)路可能是深入分析PTSD發(fā)病機(jī)理的突破口。近年來(lái),我們?cè)谶@一研究方向上取得一系列研究成果。
我們?cè)趯?shí)驗(yàn)室里建立了小鼠與天敵“戰(zhàn)斗”的行為范式,并發(fā)現(xiàn)天敵攻擊時(shí)產(chǎn)生的傷害性機(jī)械刺激是觸發(fā)小鼠戰(zhàn)斗行為的關(guān)鍵。我們?cè)谛∈蟮南虑鹉X前核 AHN中發(fā)現(xiàn)編碼傷害性機(jī)械刺激的神經(jīng)元。光激活這些神經(jīng)元,能直接引發(fā)小鼠對(duì)抗天敵的防御行為。抑制這些神經(jīng)元,小鼠失去了對(duì)抗天敵的能力。這一研究成果于2022年1月3日發(fā)表在Nature Neuroscience上。
大腦如何啟動(dòng)“逃跑”的行為反應(yīng)呢? 我們?cè)谛∈蟮拇竽X中發(fā)現(xiàn)一群?jiǎn)?dòng)逃避天敵行為的神經(jīng)元。這些神經(jīng)元位于一個(gè)叫做“上丘”的腦區(qū),以小清蛋白(parvalbumin,PV)為生物標(biāo)記物。上丘PV神經(jīng)元可以像雷達(dá)一樣監(jiān)測(cè)天空中快速逼近的運(yùn)動(dòng)目標(biāo),并把這些預(yù)警信息傳送給恐懼中樞杏仁核。直接刺激該神經(jīng)環(huán)路,可以快速引發(fā)強(qiáng)烈的逃跑行為。反復(fù)刺激該神經(jīng)環(huán)路,動(dòng)物出現(xiàn)類似PTSD的核心癥狀。這些科學(xué)發(fā)現(xiàn)于2015年6月26日發(fā)表在Science上。
以上關(guān)于“戰(zhàn)斗”和“逃跑”行為反應(yīng)的神經(jīng)機(jī)制的研究結(jié)果為進(jìn)一步深入研究PTSD的發(fā)病機(jī)理奠定了重要基礎(chǔ)。
